Blood parameter analysis and morphological alterations as biomarkers on the health of Hoplias malabaricus and Geophagus brasiliensis BABT
Romão,Silvia; Donatti,Lucélia; Freitas,Matheus O.; Teixeira,Josiane; Kusma,Josiana.
This study aimed to assess the influence of the environment on fish health. Samples of Hoplias malabaricus and Geophagus brasiliensis, were collected from three different environments: area I was urban and areas II and III were rural. Analyses of red blood cell count, microhematocrit, hemoglobin concentration, white blood cell count and differential white cell count in blood smear were carried out. Mean corpuscular volume and mean corpuscular hemoglobin concentration were calculated. To analyze morphological alterations, gills, liver, kidney and gonads were submitted to routine histological processing. Individuals collected from area III had slightly lower blood indices than collected from area I . Severe kidney changes, degeneration of and crystallization...

Storti-Filho,Agenor; Damke,Edilson; Carrara,Márcia Aparecida; Batista,Marcia Regina; Donatti,Lucélia; Boer,Cinthia Gandolfi; Svizdinski,Terezinha Inez Estivalet; Consolaro,Márcia Edilaine Lopes.
The objective of this study was to determine the effect of medroxyprogesterone acetate (DMPA) on the development and maintenance of Candida albicans in the vagina of oophorectomized Wistar rats. The animals were divided into negative control groups (NCG), which received injections of sterile saline; positive control groups (PCG), which were given injections of estradiol valerate; and progesterone groups (PG), which were given injections of Depo-Provera®. After one week of hormonal induction, vaginal infection by C. albicans was induced in all the groups and detected by vaginal yeast culture and Papanicolaou smear. In addition, scanning and transmission electron microscopy images were obtained to confirm the vaginal infection by yeast in PG. A difference in...

Effects of food deprivation in muscle structure and composition of traíra (Hoplias malabaricus): potential implications on flesh quality BABT
Rios,Flavia Sant'Anna; Donatti,Lucélia; Fernandes,Marisa Narciso; Kalinin,Ana Lucia; Rantin,Francisco Tadeu.
Hoplias malabaricus (traíra) were experimentally starved (0 to 240 days) and subsequently re-fed for 30 days (after 90 and 240-d fasting). Long-term starvation produced minor changes in muscle composition, but shrinkage of muscle fibres was a conspicuous result. The diameter of muscle fibres decreased gradually and the spaces among them increased as the starvation period progressed. After re-feeding, fish prior starved for 90 days presented partial restoration on diameter of muscle fibres. However, the fibres remained shrunken after re-feeding following 240-d fasting. While the lipid content did not change, the protein levels fell after 240-d fasting, but they were restored after re-feeding

Fine structure of the retinal pigment epithelium and cones of Antarctic fish Notohenia coriiceps Richardson in light and dark-conditions Zoologia
Donatti,Lucélia; Fanta,Edith.
The Antarctic fish Notothenia coriiceps Richardson, 1844 lives in an environment of daily and annual photic variation and retina cells have to adjust morphologically to environmental luminosity. After seven day dark or seven day light acclimation of two groups of fish, retinas were extracted and processed for light and transmission electron microscopy. In seven day dark adapted, retina pigment epithelium melanin granules were aggregated at the basal region of cells, and macrophages were seen adjacent to the apical microvilli, between the photoreceptors. In seven day light adapted epithelium, melanin granules were inside the apical microvilli of epithelial cells and macrophages were absent. The supranuclear region of cones adapted to seven day light had...

Gills of juvenile fish piaractus mesopotamicus as histological biomarkers for experimental sub-lethal contamination with the Organophosphorus Azodrin®400 BABT
Rudnicki,Cácia Aparecida Mendes; Melo,Gisele Cristiane; Donatti,Lucélia; Kawall,Helena Gonçalves; Fanta,Edith.
Piaractus mesopotamicus is a freshwater native fish from rivers of the Paraná-Paraguay Basin and of the Pantanal region and has been used for repopulation programs in Brazil. Juvenile fishes were exposed to the sub-lethal dose of 1.08mg/L of the OP Azodrin®400 containing 0.43µL/L of the active principle monocrotophos for 96 h. A frequent pathology in the gills at all times of exposure was epithelial detachment, from minimal until 24 h of exposure, to intense after 48 h of contamination. Deformed pillar cells in the respiratory lamellae leading to irregular blood spaces and blood congestion, as well as hyperplasia and lamellar fusion were observed. These histopathologies suggested that 48 h after T0 was an important time when a reduction in the capability...

Morphology of the retina in the freshwater fish Metynnis roosevelti Eigenmann (Characidae, Serrasalminae) and the effects of monochromatic red light Zoologia
Donatti,Lucélia; Fanta,Edith.
The retina of Metynnis roosevelti Eigenmann, 1915, a very active freshwater fish, was investigated by light and electron microscopy and was found to have a complex neuronal structure that allows rapid responses of visual stimuli. Retina photoreceptors are double cones, single long cones, single short cones and rods. Cone inner segments are arranged as mosaics. The outer nuclear layer contains small nuclei of twin cones, long and wide nuclei of long single cones, spherical large nuclei of short cones, and small dense nuclei of rods. Horizontal, amacrine, bipolar and ganglion neurones are responsible for connections and integration between photoreceptor cells and afferent neurones. The pigmented epithelium comprises a single layer of cylindrical cells each...

Photoperiod and feeding behavior of the Antarctic fish Notothenia rossii (Perciformes: Nototheniidae) and functional morphology of chemical and visual sensory structures used in foraging Zoologia
Donatti,Lucélia; Zaleski,Tânia; Calil,Patrícia; Fanta,Edith.
The influence of photoperiod on the ability of Notothenia rossii Richardson, 1844 to detect prey was studied experimentally. In 12L/12D there was the highest number of individuals stimulated visually and chemically while in 24D chemical perception was the same as that observed in 12L/12D, the latter being lower than in 24L. The ultrastructure of chemo- and photo-sensory structures, involved in the detection of food, was studied. The photoreceptor cells of the retina are single and double cones, and rods. Taste buds were concentrated in the central part of upper and lower lips. Their shape is even, but the size is variable. The olfactory rosette has a sensory and a non-sensory epithelium, being rich in different mucosubstances. The conclusion was that in...

Population structure and somatic indexes of Hypostomus cf. ancistroides (Siluriformes, Loricariidae) collected from the Bonito river, Ivaí river basin, Turvo, Paraná BABT
Viana,Douglas; Wollf,Luciano Lazzarini; Zaleski,Tânia; Romão,Silvia; Bertoldi,Gustavo; Donatti,Lucélia.
This study aimed to provide information about the population structure and somatic index of Hypostomus cf. ancistroides collected from the Bonito river, located in the Ivaí river basin. The length-weight relationship was isometric for both the sexes. The length structure analysis showed that the larger individuals (from 18.1cm to 27.0cm in length) predominated, and the lowest abundances occurred at the size extremes (9.1-12cm and 27.1-30.0cm). The reproduction period occured between October and January for the females and between November and January for the males. The liver somatic index cannot be used as an indicator of the reproduction period in either of the sexes, due to no correlation between the liver somatic index and the gonad somatic index. The...

Population structure of Phalloceros caudimaculatus (Hensel, 1868) (Cyprinodontiformes, Poeciliidae) collected in a brook in Guarapuava, PR BABT
Wolff,Luciano Lazzarini; Hreciuk,Ericsson Ricardo; Viana,Douglas; Zaleski,Tânia; Donatti,Lucélia.
The population structure of Phalloceros caudimaculatus was studied in a brook (sites A and B) existing in the CEDETEG - Guarapuava/PR. Fourteen length classes of 3 mm amplitude were defined for P. caudimaculatus. The widest length amplitude and the average and maximum sizes were observed in females. Females were significantly more predominant in the whole sample and more frequent in all length classes. The sexual proportion data, showed that the number of females was superior to that of males throughout most of the year. Young occurrence peaks were registered in November and March. Pregnant females were present from March to April, from July to August and in January, characterizing the reproductive season. At sampling site A, a higher occurrence of...

Retinomotor movements in the Antarctic fish Trematomus newnesi Boulenger submitted to different environmental light conditions Zoologia
Donatti,Lucélia; Fanta,Edith.
The Antarctic fish Trematomus newnesi (Boulenger, 1902) occurs from benthic to pelagic habitats, in seasonally and daily varied photic conditions that induce retinomotor movements. Fish were experimentally kept under constant darkness or light, and 12Light/12Dark for seven days. The retinomotor movement of the pigment epithelium was established through the pigment index, while that of the cones was calculated as the length of the myoid. The retinomotor movement of the pigment epithelium in T.newnesi, revealed that the adaptation to constant light occurred in the one hour of exposure, remaining constant for the next seven days. However, the adaptation to constant darkness, was slower. The difference between the mean values of the pigment indices in the time...
